Both men - aged 22 and from Sheffield - were taken to hospital, where the passenger was pronounced dead and the driver is in a stable condition. Police said they believed it was a targeted attack as the car queued at traffic lights.
No arrests have yet been made. One lane of Sheffield Parkway out of the city centre is currently shut. The second victim is currently in hospital in a stable condition. The men have not been named and there have been no arrests.
"It's anticipated they will remain closed throughout the morning as police continue their enquiries. The Parkway into the city is unaffected," a police spokesman said. South Yorkshire Police said the men were shot at from the passenger side of the car, a Silver Ford Mondeo, while the vehicle was at traffic lights near the Parkway.
Detectives have appealed for anyone who witnessed the shooting or may have information to come forward. Det Chief Insp Phil Etheridge of South Yorkshire Police said: "There is a possibility this was a targeted attack against the two victims, however it is too early in the investigation to confirm at this stage.
"I'd like to appeal to anyone who may have witnessed the incident, or anyone who has information, to please come forward and contact police."
Formal identification and a post-mortem examination are yet to take place, and no arrests have yet been made.
One lane of Sheffield Parkway leaving the city centre is closed, but a police spokesperson said the Parkway into the city was unaffected.
