Police have closed the M9, M876 and M80 approach roads.
A police spokeswoman said the road was not expected to reopen for a "considerable time".
Police said specialist "chicken catchers" and vets are at the scene.
"The lorry is blocking the carriageway and there are live chickens on the road," the spokeswoman said.
"Anyone who is trying to get on to the A80 Glasgow-bound from the M9, M876 or the M80 will be unable to do so."
