Two German people also died. Of 14 injured Britons who were taken to hospital, 13 were later released. Katherine Roberts, 50, from Pontypool, Torfaen and Karen Murray, 33, from Glasgow died with two German tourists.
The Foreign Office said the dead women were not believed to be related. All but one of 14 injured Britons taken to hospital were later released.
Their next of kin were being informed, a Foreign Office spokeswoman said. Mrs Roberts' husband Gareth, 51, remains in hospital after the crash near Seville.
The bus was on an excursion from Faro.
Police said much of the side of the bus had been ripped off on impact.
The lorry had broken down and pulled over to the hard shoulder.
Next of kin have been informed, a Foreign Office spokeswoman said.
