It follows a report, from a child, of a girl of about 10 years old with long dark hair and glasses in the water. A search that evening failed to find the teenager.
Police said they were taking the sighting seriously but no-one has been reported missing. The boy, who lives locally, had been with two friends and was in the water near the Woody Islands and North Muirton when he disappeared.
Helicopters scrambled River search
Police Scotland and the Scottish Fire and Rescue Service, supported by the ARCC helicopter from Lossiemouth, were involved in the search on Sunday evening.
The operation on Monday involved the police and fire services as well as the Police Scotland helicopter and a specialist team of police divers from Glasgow, as well as the police Search and Rescue Unit in Tayside and the civilian Tayside Mountain Rescue Team.
About 30 people will be involved in the physical search of the river, starting in the North Muirton area and heading downstream.
Searches will be carried out along the river bank and around the islands.

Officers looking for the boy said they were "carrying out a search of the River Tay along with Tayside Fire and Rescue and a search and rescue helicopter from Lossiemouth".
A spokesman added: "This is in relation to a 16-year-old local male who got into difficulties near to the Woody Island area at North Muirton, Perth, about 7.20pm and disappeared from view into the river."
