One of the workers suffered a head injury and the other a back injury in the incident.
A British Transport Police spokesman said: "The carriage that derailed was the rear carriage of a three-carriage train in the tunnel just before the SECC stop.
We expect the line will be closed for the rest of the day Network Rail
"Two members of the rail staff were in the rear carriage and suffered minor injuries."
There appeared to be no suspicious circumstances around the derailment at this stage, the spokesman added.
Officials from the Rail Accident Investigation Branch are investigating the cause of the derailment.
Network Rail said some services into Glasgow Central station were likely to be disrupted for the rest of the day while engineers inspected the scene.
A spokeswoman said: "An investigation is under way. We expect the line will be closed for the rest of the day."
