The fire service said a number of cars were involved and there had been multiple casualties. A Mitsubishi people carrier, carrying seven people including two children hit a central reservation between Sedbergh and Kirby Lonsdale, near Kendal.
The accident happened at about 1720 BST between Sedbergh and Kirby Lonsdale, near Kendal. The five other people in the car were taken to hospital and the motorway was closed on Tuesday evening in both directions until just after midnight.
The southbound carriageway between junctions 36 and 37 has reopened. The northbound route is expected to be reopened overnight. The names of the dead are being withheld until relatives are informed.
The suggested diversion for northbound traffic is to leave the M6 at Junction 36 and use the A590 and A6 north to rejoin the M6 at Junction 39. Police have appealed for any witnesses to the accident to contact them.
A police spokeswoman urged drivers to avoid the area if at all possible.
