Officers said one of the anglers had made a 999 call just after 0300 BST saying one of their group had not been seen for 20 minutes.
"There has been no sign of the man so far," said a police spokesman.
The pebble Chesil Beach is 18 miles long, and stretches from Portland to West Bay in the centre of the Jurassic Coast.
