The event, which started in 1993 with just 800 runners, starts at the city's Bellahouston Park and sees serious athletes running alongside enthusiastic amateurs. Meanwhile, Madeleine's granmother appealed to those holding their "shining star" to "take her somewhere safe" so she can be returned home.
The route makes its way round Glasgow's south side, starting and finishing in Bellahouston Park. Susan Healy told BBC News that people should look out for her granddaughter's distinctive "black flash" in her eye, where her pupil runs into her iris.
On its way it passes through the Maxwell Park and Pollok Park areas. Mrs Healy was speaking while attending the annual Liverpool 10K race, where runners were also wearing T-shirts bearing Madeleine's photo to raise awareness of the missing youngster.
A total of 64,000 bottles of water will be handed out to runners at three points along the route. Police have declined to confirm or deny reports about any possible leads, citing Portuguese law.
A spokesman for the event said: "The Women's 10K race has, over the years, become the largest women's road race in the UK with approximately 15,000 runners participating." The international number for Crimestoppers is +44 1883 731 336. People with information about Madeleine can call anonymously.
