The M27 was shut westbound at junction 11, near Fareham, and police said it would remain shut for several hours. A man in his early 20s was pronounced dead at the scene of the crash, which led to the closure of the westbound carriageway for at least eight hours.
A police spokeswoman said at least one person had died and there were "a number of casualties". A police spokeswoman said one man was arrested on suspicion of manslaughter following the accident.
Motorway re-opening
A 20-year-old man suffered life-threatening internal injuries in the crash and was taken to the Queen Alexandra Hospital in Cosham, Portsmouth.
Another man, aged 28, also sustained serious chest, back and abdominal injuries and was taken to Southampton General Hospital.
The motorway was expected to be re-opened to traffic at about 0430 GMT on Friday.
At least five vehicles crashed, just before 2000 GMT on Thursday, and police are trying to establish whether any more were involved.
Witness appeal
"One man has been arrested on suspicion of manslaughter and is currently in custody helping police with their inquiries," the spokeswoman said.
"Officers are keen to hear from anyone who may have witnessed the incident, or anyone who may know anyone who was involved in the crash."
A 32-year-old man also suffered minor injuries in the crash. All those involved in the crash are from the Southampton area.
Witnesses should contact the road death investigation team at Eastleigh police station.
