The student, believed to be aged around 19, walked into the college on Friday morning with two pistols, witnesses told Greek television.
He was said to have been heading for the classrooms, but shot a student who passed him on the staircase.
The gunman then turned to leave the school and was confronted by two older men at the gates, witnesses said.
The teenager shot at the two men before running across the road and shooting himself in the head.
"The gunman is in very serious condition with a gunshot wound to the head ... he's about to receive surgery," senior health ministry official Panayiotis Efstathiou told the Associated Press news agency.
"The victim who is seriously injured is 18 or 19 years old. The other two are about 30 and are not seriously hurt."
Our correspondent in Athens said it was not clear if the gunman was a student at the college, which offers vocational training for the unemployed.
