South Wales Police said the victim came from Tregelles Court in the Cwmrhydyceirw area of Morriston.
He was driven to Lasau Road near Junction 46 of the M4.
Det Insp Simon Davies said: "This is being treated as an isolated incident and we are seeking the whereabouts of a 35-year-old man from the Gendros area, and as yet two unidentified males.
"The victim has been left traumatised by the unprovoked actions of these individuals and I am appealing for anyone who witnessed anything untoward at Tregelles Court or Pant Lasau Road between 0200 and 0300 GMT to come forward."
